Mercaptopurine inhibits purine nucleotide synthesis and metabolism by inhibiting phosphoribose pyrophosphate amide transferase (PRPP amide transferase). PRPP amide transferase is a rate-limiting enzyme formed by purine. It alters the synthesis and function of RNA and DNA. Mercaptopurine also interferes with nucleoside tautomies and glycoprotein synthesis.
