Biological Activity

MQAE is a novel chloride ion fluorescence probe excited by ultraviolet light. MQAE, binding bromine ion as ligand, can be used to detect the activity of chloride. MQAE exhibits a reduction of fluorescence intensity with the increase of intracellular Cl- concentration. MQAE has a pH-independent fluorescence intensity basically at a Cl- concentration <50 nM. MQAE has high water solubility and cell permeability, MQAE has been used for fluorescence detection such as confocal microscopy and flow cytometry. MQAE is a fluorescent indicator that is quenched via collision with chloride, and is more sensitive and selective than 36Cl and microelectrode-based methods for chloride measurement in cells. For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
