Biological Activity

KN-93 hydrochloride is a cell-permeable inhibitor of Ca2+/calmodulin-dependent protein kinase II (CaMK-II) with a Ki of 370 nM. KN-93 HCl inhibits fibroblast CaMK-II activity and cell growth in a dose-dependent manner, reversibly arrests cells in G1 and induces apoptosis. KN-93 also blocks cell growth stimulated by basic fibroblast growth factor, platelet-derived growth factor-BB, and epidermal growth factor in NIH 3T3 fibroblasts. KN-93 (0.5 μM) prevents increased LV developed pressure during action potential prolongation and early afterdepolarizations.

Animal A (mg/kg) = Animal B (mg/kg) multiplied by  Animal B Km
Animal A Km

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
