Biological Activity

Tormentic Acid is a natural anti-inflammatory agent which inhibits production of tumor necrosis factor-α (TNF-α), interleukin 6 (IL-6), and IL-1β. Tormentic Acid suppresses the NF-κB signaling pathway. Tormentic acid has anti-inflammatory activity, it potently inhibits the production of nitric oxide (NO) in RAW 264.7 cells, also suppresses the LPS-stimulated degradation and phosphorylation of inhibitor of kappa B-α (IκB-α), suggests that the anti-inflammatory activity of TA is associated with the down-regulation of iNOS, COX-2, and TNF-α through the negative regulation of the NF-κB pathway in RAW 264.7 cells. Tormentic acid has protective effect against lipopolysaccharide/D-galactosamine induced fulminant hepatic failure in mice.

Animal A (mg/kg) = Animal B (mg/kg) multiplied by  Animal B Km
Animal A Km

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
