R-Spondin-1 (Rspo-1) belongs to the Wnt regulator (Rspo) family. Currently, the family consists of four structurally related secretory ligands (Rspo 1-4), all containing basic residues and thrombospondin structural domains. R-Spondin-1 is a secreted activator protein containing two adjacent cysteine furan domains (aa 34-135) with a potential N-glycosylation site, followed by a thrombospondin (TSP1) motif (aa 147-207) and a region rich in basic residues (aa 211-263). Rspo-1 is expressed in certain regions of the developing central nervous system, as well as the adrenal gland, ovary, testis, thyroid and trachea. Rspo can interact with the Frizzled/LRP6 receptor complex to stimulate the Wnt/β-catenin signaling pathway.
