Biological Activity

Recombinant Human Noggin Protein (HEK293) can bind to ligands of the transforming growth factor-β family and regulate its activity by inhibiting its pathway with signal receptors. Noggin was initially identified as a BMP-4 antagonist, and its effect is essential for the normal formation of the head and other back structures. Therefore, noggin has been shown to regulate the activities of other BMP-2, -7, -13 and -14. Transgenic mice overexpress noggin in mature osteoblasts, which is manifested by impaired differentiation of osteoblasts, decreased bone formation, and severe osteoporosis.

Determined by the ability of ATDC chondrocytes to inhibit the production of alkaline phosphatase by 5.0 ng/ml BMP-4, the expected ED50 of this effect is about 2.0-3.0 ng/ml.

The DNA sequence encoding human noggin (NP_005441.1) is expressed with a His tag at the C terminus.

Recombinant human noggin contains 211 amino acids and has a predicted theoretical molecular weight of 23.9 kd.
