Biological Activity

Dipeptidyl peptidase-4 (DPP4) is a serine exopeptidase belonging to the S9B protein family. The enzyme is a type II transmembrane glycoprotein, expressed on the surface of many cell types. It is also present in serum and other body fluids in a truncated form (sCD26/DPPIV). As both a regulatory enzyme and a signalling factor, DPP4 has been evaluated and described in many studies. DPP4 inhibition results in increased blood concentration of the incretin hormones glucagon-like peptide-1 (GLP-1) and gastric inhibitory polypeptide (GIP).

The recombinant human DPP-4 expressed the sequence encoding the extracellular domain (Asn 29-Pro 766) of the mature form of human DPPIV with the fused Fc region of human IgG1 at the N-terminus.

Measured by its ability to cleave the fluorogenic peptide substrate, Gly-Pro-7-amido-4-methylcoumarin, the specific activity is > 2000 pmoles/min/μg.

DPP4/CD26 is a signal-anchor for type II membrane protein that belongs to the peptidase S9B family. DPP4/CD26 acts as a positive regulator of T-cell coactivation, by binding at least ADA, CAV1, IGF2R, and PTPRC. It’s binding to CAV1 and CARD11 induces T-cell proliferation and NF-kappa-B activation in a T-cell receptor/CD3-dependent manner.
