PAMAM-NH2 TG64 is a polymer with an ethylenediamine core that forms a highly branched dendritic structure; it is widely used in fields such as drug delivery, gene transfer and bioimaging. This structure provides a larger surface area and greater functionality, facilitating the efficient packing of DNA/RNA within the core region and promoting cell membrane destabilisation, thereby demonstrating excellent performance in gene transfection. The terminal amino groups provide abundant reactive sites capable of cross-linking with a variety of biomolecules or drugs, thereby enhancing its chemical reactivity.
Applications
1. Gene/nucleic acid delivery: PAMAM is one of the most established non-viral gene delivery vectors; it forms electrostatic complexes with the phosphate backbone of nucleic acids via surface amino groups, thereby mediating the intracellular delivery of DNA, siRNA and mRNA
2. Drug Delivery: As a nanocarriage for drugs, it can load both hydrophobic and hydrophilic drugs, whilst its surface amine groups facilitate further modification with targeting ligands (such as folic acid, transferrin, PEG, etc.)
3. Material modification: As a processing aid for polyamides (nylon), it improves melt flow, heat resistance and hydrolysis resistance.
4. Nanoparticle stabilisation: By utilising the high density of surface amine groups to interact with the surface of nanoparticles, it provides dispersion stabilisation and functional modification.
