Biological Activity

DOTMA, as a tetra-methylated DOTA analogue, is a cationic lipid and can be used as a non-viral vector for gene therapy. It has been used as a component in liposomes that can be used to encapsulate siRNA, microRNAs, and oligonucleotides and for gene transfection in vitro. It exhibits effective in vitro and in vivo gene transfection. DOTMA is applied to DNA delivery. Studies have found that single-compartment liposomes prepared by DOTMA can encapsulate DNA and RNA fragments at a high level, with an encapsulation rate of 100%, and can be well taken up by cells to express target genes. The transfection efficiency at the cellular level is 10 times higher than that of DOTAP. The prescription is also simpler, and there is no need to add helper lipids such as DOPE.

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
