Benpyrine racemate ((Rac)-Benpyrine) is a racemate of Benpyrine, Benpyrine racemate ((Rac)-Benpyrine) is a potent and orally active TNF-α inhibitor and has the potential for TNF-α mediated inflammatory and autoimmune disease research. (Rac)-Benpyrine (5-10 μM; 24 hours) treatment inhibits the expression level of TNF-α downstream pathway protein NF-κB in the nucleus in RAW264.7 macrophages. In vivo, (Rac)-Benpyrine (25-50 mg/kg/day; oral gavage; 3 weeks) treatment obviously reduces the inflammatory cytokines IFN-γ, IL-1β and IL-6 in the blood serum, and the anti-inflammatory factor IL-10 is obviously increased, the foot swelling degree of the arthritis part is also obviously slowed down.

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
