Biological activity: CD20 is a specific marker molecule on the surface of B lymphocytes. It is expressed on more than 95% of normal or malignant B lymphocytes and is not expressed in hematopoietic stem cells, progenitor cells, and other normal tissues. It is composed of 297 amino acids and has a molecular weight of 33kD. It belongs to non-glycosylated phosphoprotein from 34 to 36kD. Its antigenic epitope is a ring region composed of 43 amino acids in the third and fourth transmembrane regions. CD20 is an ideal antigen for the treatment of B-lymphocyt-related diseases because it has no obvious internalization and no obvious cell surface shedding after binding with antibody. Since the original normal B cells were not affected by the anti-CD20 monoclonal antibody, the anti-CD20 monoclonal antibody was able to rebuild the B cell population after killing most, if not all, lymphoma cells expressing the CD20 molecule as well as normal B cells. Therefore, anti-CD20 monoclonal antibodies are increasingly used in the treatment of B-cell related diseases.
