Biological Activity

Sonic Hedgehog Homolog (SHH) belongs to the Hedgehog family of three proteins. The other two family members are the Indian hedgehog (IHH) and desert hedgehog (DHH). Hedgehog proteins are key signaling molecules for embryonic development. SHH is expressed in a variety of embryonic tissues and plays a key role in regulating the patterns of many systems, from limbs to the brain. SHH also plays an important role in adults, including the division of adult stem cells and the development of certain cancers and other diseases. Human SHH is expressed as a 45kDa precursor and undergoes a series of processes during secretion. After the removal of the signal peptide, a protease located in the C-terminal domain catalyzes the cleavage of SHH into a 20 kDa N-terminal domain (SHH-N) and a 25 kDa C-terminal domain (SHH-C). Shh-n has "full signal" capability. Shh-n binds to Patched 12-channel transmembrane proteins (Ptc) on the cell surface to release Ptc's inhibitory effect on THE activity of G protein-coupled receptor Smoothened (Smo).
