M3554 is a novel anti-GD2 ADC designed based on humanized ch14.18 anti-GD2 antibody. M3554 carries the topoisomerase I inhibitor (TOP1i) exatecan as a payload, linked by a cleavable β-glucuronide ligand. M3554 exhibits antitumor activity in vivo, and results in tumor regression in the CHP134 xenograft model. In addition, M3554 showed good PK and a favorable safety profile in rats and monkeys.
