Biological Activity

Endothelin-1 (human, porcine) is a synthetic peptide with the sequence of human and swine Endothelin 1, which is a potent endogenous vasoconstrictor. Endothelin-1 acts through two types of receptors ETA and ETB. ET-1 causes fibrosis of the vascular cells and stimulates production of reactive oxygen species. It is claimed that ET-1 induces proinflammatory mechanisms, increasing superoxide anion production and cytokine secretion.

Preparation method Cardiomyocytes were cultured in 24-well culture plates. Twelve hours after the addition of rotenone (1 μM), endothelin-1 (10 nM) or vehicle was added to the medium. Concomitantly, 37 kBq of 2-deoxyglucose solution was added to the culture medium. The cells were incubated for 7 h. Then, the medium was aspirated, and the cells were washed twice with ice-cold phosphate-buffered saline.
