Biological Activity

Diclofenac is a non-selective COX inhibitor with IC50 of 60 and 220 nM for ovine COX-1 and -2, respectively. It inhibits human COX-1 and -2 with IC50 values of 0.9-2.7 and 1.5-20 uM, respectively. It is thought that the primary mechanism responsible for its anti-inflammatory, antipyretic, and analgesic action is inhibition of prostaglandin synthesis by inhibition of cyclooxygenase (COX), and it appears to inhibit DNA synthesis.

Animal A (mg/kg) = Animal B (mg/kg) multiplied by  Animal B Km
Animal A Km

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
