Bioactivity: CD3/CD28 mab conjugated magnetic beads are mainly used for isolation, activation and in vitro amplification of human T cells. The use of 4.5-um superparamagnetic beads, matched to cell size, coupled with anti-CD3 and CD28 antibodies, provides the primary and co-stimulatory signals required for T cell activation and amplification. First, CD3/CD28 immunomagnetic beads were used for magnetic cell isolation, and CD3+ T cells were isolated and enriched from the obtained isolated products. After isolation, CD3+ T cells were cultured in the presence of magnetic beads. By binding anti-CD3 and anti-CD28 antibodies on immunomagnetic beads, the beads provide primary and costimulatory signals required for T cell activation and amplification. Activated T cells produce IL-2 (interleukin 2), GM-CSF (granulocyte macrophage stimulating factor), IFN-γ (interferon γ) and TNF-α (tumor necrosis factor α), which play the role and function of T cells.

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
