Ionomycin causes a rapid rise in cytosolic Ca2+ that is due to both a release of cytosolic Ca2+ stores and Ca2+ influx. Ionomycin also activates (2 microM) and primes (20-200 nM) the PMN oxidase. Ionomycin is shown to be an effective ionophore for Ca2+ in rat liver mitochondria. DNA and protein analysis in Ionomycin (SQ23377)-treated cultures revealed DNA fragmentation and PARP cleavage to an 85-kDa fragment typical of caspase-mediated apoptosis. Three hours after addition of ionomycin, the percentage of early apoptotic cells approximately doubles. Necrosis could be detected in ~1-5% of the Ionomycin (SQ23377) treated cells as supported by simultaneously positive fluorescein labeling and propidium iodide uptake.

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
