Maprotiline hydrochloride is a selective noradrenalin re-uptake inhibitor and a tetracyclic antidepressant. Maprotiline hydrochloride acts primarily by potentiation of central adrenergic synapses by blocking reuptake of norepinephrine at nerve endings. Maprotiline hydrochloride is indicated for the treatment of depressive illness in patients with depressive neurosis (dysthymic disorder) and manic-depressive illness, depressed type (major depressive disorder). Maprotiline hydrochloride is also effective for the relief of anxiety associated with depression.

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.

CRN04894
CRN04894 is an orally nonpeptide, potent and selective antagonist for adrenocorticotropic hormone (ACTH) acting at the melanocortin 2 receptor (MC2R). CRN04894 blocks the action of ACTH at MC2R, providing a potential novel treatment for the diseases of ACTH excess such as Cushing’s disease, congenital adrenal hyperplasia, and ectopic ACTH-secreting tumors. |
