Biological Activity

Leonurine hydrochloride is an alkaloid isolated from Herba leonuri, with anti-oxidative and anti-inflammatory.

Customer Product Validations & Biological Datas
Source Bone (2015). Figure 2. Leonurine-hydrochloride
Method qRT-PCR
Cell Lines RAW264.7 cells
Concentrations 2 μM
Incubation Time 24 h
Results The mRNA expression of RANK was upregulated in M-CSF treated BMMs. LH treatment significantly suppressed RANK mRNA expression in BMMs
Source Bone (2015). Figure 1. Leonurine-hydrochloride
Method osteoclastogenesis assay
Cell Lines RAW264.7 cells
Concentrations 2 μM
Incubation Time 5 d
Results LH inhibited osteoclastogenesis in both RAW 264.7 cells (Fig. 1A) and BMMs (Fig. 1B) in a dose-dependent manner from 0.5–2 μM.
Protocol (for reference only)
Cell Experiment
Cell lines RAW 264.7 cells
Preparation method RAW 264.7 cells were seeded in a 6-cm dish and pretreated with different concentrations of LH (2 μM) for 6 h, and then stimulated with RANKL (100 ng/mL) for 15 min. The cells were washed with icecold PBS and lysed in radioimmunoprecipitation assay (RIPA) buffer.
Concentrations 2 μM
Incubation time 6 h
Animal Experiment
Animal models Male Sprague-Dawley rats
Formulation 0.5% sodium carboxymethyl cellulose (CMC) solution
Dosages 30 mg/kg
Administration oral
Chemical Information
Molecular Weight 347.79
Formula C14H21O5N3.HCl
CAS Number 24735-18-0
Solubility (25°C) Water 4 mg/mL
DMSO 24 mg/mL
Storage 4°C, dry, sealed

Animal A (mg/kg) = Animal B (mg/kg) multiplied by  Animal B Km
Animal A Km

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
