Biological Activity

Actinomycin D is a compound composed of a two CYCLIC PEPTIDES attached to a phenoxazine that is derived from STREPTOMYCES parvullus. It binds to DNA and inhibits RNA synthesis (transcription), with chain elongation more sensitive than initiation, termination, or release. As a result of impaired mRNA production, protein synthesis also declines after dactinomycin therapy.

Method RNA decay assay
Cell Lines MHCC97H and HepG2 cells
Concentrations 5 μg/mL
Incubation Time -
Results We next sought to assess the RNA decay rate in Mettl3-WT and Mettl3-KR cells treated with the transcription inhibitor actinomycin D to block new RNA synthesis. Snail mRNA expression was decreased, and the Snail mRNA half-lives were significantly shortened in SUMO-mutant Mettl3 cells

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
