IL-23 (IL-23 alpha & IL-12 beta Heterodimer) Protein is a recombinant protein dimer complex containing rat-derived IL-23 alpha & IL-12 beta Heterodimer protein, expressed by HEK293. The IL-23 alpha protein has cytokine activity and binds to interleukin-23 receptors. It regulates cytokine production, lymphocyte activation, and peptidyl-tyrosine phosphorylation. It influences T cell proliferation and RNA polymerase II transcription. Found in the extracellular space, it is part of the interleukin-23 complex. It is mainly expressed in the thymus, spleen, and other tissues. It is orthologous to the human IL23A gene encoding interleukin 23 subunit alpha.
The recombinant rat IL23A & the rat IL12B comprises 510 (186+324) amino acids and has a calculated molecular mass of 58.2 (21+37.2) KDa. A DNA sequence encoding the rat IL23A (Met1-Ala196) was fused with a polyhistidine tag at the C-terminus, constructed the plasmid 1; A DNA sequence encoding the rat IL12B (Met1-Ser335) was expressed with a polyhistidine tag at the C-terminus, constructed the plasmid 2. The two plasmids were co-expressed and the rat IL23A & the rat IL12B heterodimer was purified.
Measured by its ability to induce IL17 secretion by mouse splenocytes, the ED50 for this effect is 0.5-3ng/mL.

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
