Wnt-3a is one of 19 vertebrate members of the Wingless-type MMTV integration site (Wnt) family. WNT signaling plays key roles in the control of development, homeostasis, and regeneration of many essential organs and tissues. Endogenous WNTs bind to multiple FZDs and are heavily modified posttranslationally, making them difficult to manufacture consistently. WNT surrogate molecules phenocopied endogenous ligand activities in in vitro reporter assays and intestine organoid functional assays.
Measured by its ability to induce Topflash reporter activity in HEK293T human embryonic kidney cells, the ED50 for this effect is 71.45 ng/ml.

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
