Solamargine is a major steroidal alkaloid glycoside extracted from a traditional Chinese medicinal herb, Solanum nigrum L. (SNL); has been shown to inhibit growth and induce apoptosis of various cancer cells.

Preparation method | Cell viability assay Cell viability was measured using the 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Briefly, lung cancer cells were harvested and seeded in a 96-well microtiter plate followed by treating with solamargine (6 μM) for up to 48 hrs. Afterwards, MTT solution (20 μl, 5 g/l) was added to each well, and cells were incubated at 37°C for an additional 4 hrs. Finally, the 200-μl solvent dimethyl sul-foxide was added to each well for 10 min. The ELISA reader (Perkin Elmer, Victor X5, USA) was used to detect the Absorbance at 490 nm. Cell viability (%) was calculated as follows: (absorbance of test sample/ absorbance of control) 100%. |
