N-Nitrosodiethylamine (NDEA) belongs to the nitrosamines category of nitrogenous disinfection byproduct. N-Nitrosodiethylamine has been used to induce hepatocarcinogenesis in rat and mice. NDMA is acted upon by the cytochrome P450 system resulting in the formation of carcinogenic methyldiazonium ion. N-Nitrosodiethylamine is responsible for the changes in the nuclear enzymes associated with DNA repair/replication. N-Nitrosodiethylamine results in various tumors in all animal species. The main target organs are the nasal cavity, trachea, lung, esophagus and liver.

For example, to modify the dose of Compound A used for a mouse (20 mg/kg) to a dose based on the BSA for a rat, multiply 20 mg/kg by the Km factor for a mouse and then divide by the Km factor for a rat. This calculation results in a rat equivalent dose for Compound A of 10 mg/kg.
