Biological Activity

Kaempferol 3-O-β-D-glucuronide is a conjugated kaempferol metabolite, which has anti-inflammatory effect. Kaempferol 3-O-β-D-glucuronide significantly inhibits various pro-inflammatory mediators like IL-1β, NO, PGE2, and LTB4.

Cell Experiment
Cell lines RAW 264.7 cells
Preparation method RAW 264.7 cells (2×105cells/well) were pretreated with K3G (5 and 10 μM) for 1 h and stimulated with LPS (1 μg/mL) for 24 h. Cells were then harvested. Cell pellet was solubilised in RIPA buffer (Sigma; R-0278) over ice for 40 min. The lysate was centrifuged at 14,000 g for 15 min at 4 °C. Supernatant was collected. Protein estimation was done by Bradford's method with BSA standard. Western blotting was performed according to the reported protocol. Detection of specific proteins was done by Immobilon Western (Chemiluminescent HRP substrate, Millipore) mediated chemiluminescence by ChemiDoc XRS+(Bio Rad). Densitometry of specific bands was done using Image Lab Software version 3 (Bio Rad). The effect on phosphorylation of NF-kB p65 was studied in a dose dependent manner in comparison to dexamethasone (0.5 μM).
